Overview:
Sermorelin is a synthetic peptide composed of 29 amino acids. It corresponds to the amino-terminal 1–29 segment of naturally occurring human growth hormone-releasing hormone, or GHRH. This portion of GHRH retains the biological activity needed to stimulate growth hormone release from the anterior pituitary gland.
Scientific and clinical interest in sermorelin centers on its interaction with the growth hormone–releasing hormone receptor and the downstream regulation of endogenous growth hormone secretion. Unlike administering growth hormone directly, sermorelin acts upstream by signaling functioning pituitary somatotroph cells to release the body’s own growth hormone.
Sermorelin has been studied in connection with pituitary function, growth hormone secretion, growth-related signaling and the evaluation of growth hormone deficiency.
Mechanism Overview:
Sermorelin binds to and activates GHRH receptors located on somatotroph cells in the anterior pituitary. This activates intracellular signaling pathways that promote the synthesis and release of growth hormone.
Released growth hormone can subsequently stimulate hepatic and peripheral production of insulin-like growth factor 1, or IGF-1, which participates in growth, metabolism and tissue-related signaling. Sermorelin’s effects depend on the functional capacity of the hypothalamic-pituitary growth hormone axis.
